目的:了解甲状腺激素受体β△基因在大鼠不同组织中的分布特点,了解实时荧光定量PCR的原理及使用方法。熟悉采用实时荧光定量PCR的方法分析结果。方法:TRβ△基因在多种组织中均有表达,其中肝脏、脑、骨胳肌、心脏、肾脏表达最多。提取大鼠肝脏、脑、骨胳肌、心脏、肾脏等多处组织的mRNA, 以提取的mRNA为模板逆转录合成cDNA第一链。并以之为模板PCR扩增成双链cDNA。根据转录出的目的基因设计两条引物。采用实时荧光定量PCR的方法进行定量分析。结果:1.提取组织总RNA,OD260/OD280均介于1.7-2.0之间,OD260/230介于1.5-1.9之间。经甲醛变性琼脂糖凝胶电泳,28S与18S的密度比值约2。2.琼脂糖凝胶电泳鉴定PCR扩增结果:1)GAPDH PCR产物的琼脂糖凝胶电泳鉴定结果显示:在预期的约160bp左右有目的扩增产物带。2)TRβΔ PCR产物的琼脂糖凝胶电泳鉴定结果显示:经PCR扩增出的DNA分子量大小为176bp的目的条带。3)重组克隆质粒pGEM-基因的鉴定结果:以EcoRⅠ对重组质粒进行酶切,2%琼脂糖凝胶电泳检测酶切结果,可以看到在3000bp左右的质粒片段,以及与插入序列大小相符的另一条DNA的条带。目的基因及管家基因定量测定,其扩增曲线和溶解曲线未见杂峰,同时融解曲主峰对应的Tm值与扩增产物的理论Tm相近。结论:研究结果显示TRβ△基因于mRNA水平的表达有一定的空间分布特点:肝,脑,脾,肾组织中均有该基因表达,且脾中的表达量明显高于其他组织。
