目的：了解甲状腺激素受体β△基因在大鼠不同组织中的分布特点，了解实时荧光定量PCR的原理及使用方法。熟悉采用实时荧光定量PCR的方法分析结果。方法：TRβ△基因在多种组织中均有表达，其中肝脏、脑、骨胳肌、心脏、肾脏表达最多。提取大鼠肝脏、脑、骨胳肌、心脏、肾脏等多处组织的mRNA, 以提取的mRNA为模板逆转录合成cDNA第一链。并以之为模板PCR扩增成双链cDNA。根据转录出的目的基因设计两条引物。采用实时荧光定量PCR的方法进行定量分析。结果：1.提取组织总RNA，OD260/OD280均介于1.7-2.0之间，OD260/230介于1.5-1.9之间。经甲醛变性琼脂糖凝胶电泳，28S与18S的密度比值约2。2.琼脂糖凝胶电泳鉴定PCR扩增结果：1）GAPDH PCR产物的琼脂糖凝胶电泳鉴定结果显示：在预期的约160bp左右有目的扩增产物带。2）TRβΔ PCR产物的琼脂糖凝胶电泳鉴定结果显示：经PCR扩增出的DNA分子量大小为176bp的目的条带。3）重组克隆质粒pGEM-基因的鉴定结果：以EcoRⅠ对重组质粒进行酶切，2％琼脂糖凝胶电泳检测酶切结果，可以看到在3000bp左右的质粒片段，以及与插入序列大小相符的另一条DNA的条带。目的基因及管家基因定量测定，其扩增曲线和溶解曲线未见杂峰，同时融解曲主峰对应的Tm值与扩增产物的理论Tm相近。结论：研究结果显示TRβ△基因于mRNA水平的表达有一定的空间分布特点：肝，脑，脾，肾组织中均有该基因表达，且脾中的表达量明显高于其他组织。
